摘要
目的 探讨经皮穿刺鞘内注射给药方法在大鼠基础实验研究中的可行性。 方法 大鼠取俯卧位 ,实验者左手的拇指和中指置于大鼠脊柱 L5- 6 间隙的两侧 ,以食指定位 ,右手持微量注射器从间隙垂直缓慢进针 ,以尾巴出现颤动或突然的侧向甩动作为穿刺成功的标志 ,注入药物。以此方法观察大鼠在给予布比卡因后的行为变化、给予造影剂后的 X线表现以及鞘内注射吗啡对炎性疼痛模型大鼠热痛缩腿反应潜伏期的影响。 结果 大鼠鞘内注射布比卡因后双后足无力且钳夹时不引起缩腿反应 ;给予造影剂的大鼠在相当于蛛网膜下腔处出现一不透 X线的条带 ;鞘内注射吗啡能明显延长炎性疼痛模型大鼠的热痛缩腿反应潜伏期。 结论 大鼠鞘内直接穿刺注射给药具有简单易行、创伤小。
Objective To explore the feasibility of a method performed via direct transcutaneous intrathecal(i.t) drug injection in the basic research of pain in rats. Methods Between L-5 and L-6 with the left forefinger, a minimal syringe punctured slowly and vertically clearance. Drugs might be injected when a flicker or a sudden lateral movement of the tail was observed, which was the sign of successful puncture. Then the method was used to observe the changes of behavior when bupivacaine was administered i.t. The changes of paw withdrawal latency(PWL) in a rat model of inflammatory pain when morphine was administered i.t. Results Bupivacaine i.t made the rat's feet disable and no response to the stimulation nipped by pliers. Intrathecal morphine increased distinctly PWL in the model of inflammatory pain. Conclusion The method of direct intrathecal injections is simply, easy to operate, and little hurt to rats. It's worthy in the basic studies of algesia and analgesia.;
