摘要
目的研究氟代柠檬酸(fluorocitrate,FC)对骨癌痛大鼠CB2基因表达的影响及意义。方法 40只SD大鼠注射Walker-256肿瘤细胞,建立骨癌痛模型,另取10只SD大鼠作为假手术组,10只作为对照组。首先比较假手术组和骨癌痛大鼠热缩足反射潜伏期(thermal withdrawl latency,TWL)。再将骨癌痛大鼠随机分为FC组和非FC组,每组20只。FC组术后14 d注射氟代柠檬酸0.01 m L(1 nmol),非FC组注射生理盐水0.01m L,记录大鼠机械缩足阈值(paw withdrawl mechanical threshold,PWMT)。同时采用免疫印迹法测定CB2表达结果,观察灰度值。结果骨癌痛组大鼠TWL先缩短后恢复正常,假手术组基本保持不变,2者在第3 d具有统计学差异(P<0.05)。FC组大鼠注射氟代柠檬酸后,PWMT值有先升高后降低现象。假手术组和骨癌痛组大鼠CB2均明显高于基础值(P<0.05);非FC组大鼠CB2逐渐降低,第7天和第14天时显著高于假手术组(P<0.05)。FC组大鼠与非FC组相比,CB2在7d时有统计学差异(P<0.05),14 d和21 d时略低,但差异无统计学意义。结论患骨癌痛后,机体CB2表达会有一定程度增加,引起疼痛,而氟代柠檬酸可抑制CB2的表达,从而缓解疼痛。
Objective To study effect of fluorocitrate on expression of CB2 in a rat model of bone cancer pain. Methods The Walker-256 tumor cells were injected into 40 rats to establish the bone cancer pain model. Another 10 SD rats were as sham operation group,and 10 rats were as control group. Compared TWL between sham operation( sham) group and bone cancer pain( BCP) group,the bone cancer pain rats were divided into FC group and non FC group,0. 01 m L( 1nmol) fluorocitrate were injected into FC group,while physiological saline were given to none FC group. Paw withdrawl mechanical threshold was recorded in rats. CB2 was determined by Western blot at the same time. Results TWL of BCP group initially decreased and then returned to normal,and sham group remained unchanged. After injection of fluorocitrate,PWMT of FC group increased. CB2 of sham group and BCP group were significantly higher than basal values( P〈0. 05). For the none FC group rats,CB2 gradually decreased,which was significantly higher than that in sham group in 7 d and 14 d( P〈0. 05). Morever,compared with the non FC group,CB2 of FC group were significantly less in 7 d( P〈0. 05).Conclusion After suffering bone cancer pain,CB2 expression may increase to a certain extent,leading to the pain. Fluorocitrate can inhibit expression of CB2,thus relieve pain.
