响应曲面法优化文冠果种仁蛋白的碱溶酸沉提取工艺 被引量:23

摘要 以文冠果种仁脱脂粉为原料,采用碱溶酸沉法提取文冠果蛋白,利用响应曲面法优化其提取工艺。运用Box-Behnken实验设计方法,在单因素实验的基础上,以蛋白质得率为实验指标,研究液料比、pH、提取时间以及提取温度等影响因素及其交互作用对蛋白质得率的影响。实验结果表明,文冠果蛋白质碱溶酸沉的最佳提取工艺参数为:液料比11∶1(mL/g),pH11,提取时间73min,提取温度41℃。在本实验范围内各影响因素对蛋白质提取率作用的大小依次为:pH>液料比>时间>温度。在此条件下,蛋白质提取率达到83.58%,利用响应面分析法得到的二次多项式回归方程数学模型Y=82.64+3.98A+3.56B+3.04C+5.11BC-4.40A2-6.63B2-5.89C2-6.79D2能较准确地预测实验结果。 The Xanthoceras sorbifolia bunge kernel protein was extracted by Alkali-Solution and Acid-Isolation.The extraction processing was optimized through response surface analysis.By use of the Box-Behnken experimental design method,the effects of the ratio of liquid,pH,extraction time and temperature and their interactions on the protein yield were studied on the base of the single-factor test.The test results showed that the optimum conditions of alkali-solution and acid-isolation extraction for Xanthoceras sorbifolia bunge protein were as following: the ratio of liquid 11∶1( mL/g) ,pH11,extraction time 73min,extraction temperature 41℃.The influence order of factors to protein extracting rate is pH ratio between solid and liquid reaction time reaction temperature.Under these conditions,the rate of protein extraction reached 83.58% . The mathematic model of the quadratic polynomial regression equation was obtained through the method of response surface analysis which could predict the yield of protein exactly.
