摘要
目的:观察自然衰老大鼠下丘脑-垂体-肾上腺(HPA)轴功能变化,并通过右归丸干预,探索补肾方延缓衰老的机理。方法:以自然衰老SD雄性大鼠(24月龄)为肾虚衰老模型,大鼠随机分为老年对照组和右归丸组,每组35只,另设青年对照组(5月龄)35只,共3组。老年对照组大鼠常规喂养至24月龄。右归丸组大鼠自20月龄始,按相当于右归丸生药量7.5g/kg自由饮用稀释药液,每周停药2天,连续4个月;采用放射性配体结合分析法观察大鼠血中促肾上腺皮质激素(ACTH)和皮质酮(CORT)含量;反转录聚合酶链式反应(RT-PCR)和蛋白印迹(Western Blot)技术观察大鼠下丘脑促肾上腺皮质激素释放激素(CRH)mRNA及其多肽表达。结果:与青年组比较,老年组大鼠下丘脑CRHmRNA和CRH表达、血浆ACTH及血清CORT含量升高;与老年组比较,右归丸组大鼠下丘脑CRHmRNA和CRH表达、血浆ACTH及血清CORT含量显著下降(P<0.05)。结论:自然衰老大鼠神经内分泌调控紊乱,HPA轴功能亢进;右归丸可调节HPA轴,使亢进的HPA轴功能得以改善,减轻对机体的危害而延缓衰老。
Objective:To study the antiaging mechanism of kidney tonifying prescriptions through observing the change of function of hypothalamus-pituitary-adrenal axis in aged rats and the action of Yougui pill.Methods:natural aging rats was as Kidney abscence model.Male SD rats were divided into three groups:young group(5 months old)、old group(24 months old,generally raised)and Yougui Pill group.From 20 months the Yougui Pill group were given Yougui Pill with 7.5g/kg for 4 months(two days with no pill every week).RIA was used to detect ACTH and CORT in blood and RT-PCR and Western-blot were used to detect the expression of CRH mRNA and CRH in hypothalamus in all groups.Results:Compared with young control group,the expression of CRH mRNA and CRH in hypothalamus and the quantity of ACTH and CORT in blood were all increased in old group.Meanwhile the expression of CRH mRNA and CRH in hypothalamus and the quantity ACTH and CORT in blood were all decreased in Yougui Pill group.Conclusion:The regulation of neuro-endocrine is disordered in natural aged rats and hyperfunction of HPA axis occurs;Yougui Pill can antiaging through eliminating the hyperfunction of HPA axis.
