补肾方药对D-半乳糖致衰大鼠心肌组织氧化应激和细胞凋亡的影响

Effects of Kidney-nourishing Herbs on Oxidative Stress and Apoptosis in Aging Rat Myocardium Induced by D-Galactose

摘要 目的:观察补肾方药对D-半乳糖致衰大鼠心肌组织氧化应激及细胞凋亡的影响。方法:2 months雄性SD大鼠40只,随机分为青年对照组、衰老组、左归丸组、右归丸组,每组10只。衰老组每日腹腔注射D-半乳糖400mg·kg-1,连续60天;青年对照组每日腹腔注射等体积生理盐水;左归丸组和右归丸组处理同衰老组,同时分别按每日相当生药量4.97g·kg-1和4.94g·kg-1灌胃给药。采用黄嘌呤氧化酶法和硫代巴比妥酸(TBA)法检测心肌组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;采用Western Blot检测Bcl-2和Bax蛋白表达水平并测定组织Caspase-3相对活性。结果:与青年组对照组相比,D-半乳糖致衰组大鼠心肌组织SOD活性下降、MDA含量升高,Bcl-2蛋白表达下调、Bax蛋白表达上调,Caspase-3活性升高;给予左归丸、右归丸能显著改善D-半乳糖致衰大鼠心肌组织氧化应激水平,减少细胞凋亡。结论:补肾方药左归丸、右归丸可通过提高抗氧化能力和减少细胞凋亡而发挥其延缓心肌组织衰老的作用。 Objective : To investigate the effects of kidney-nourishing herbs on oxidative stress and apoptosis in aging rat myocardium induced by D-Galactose. Methods :40 male SD (2 months)rats were equally divided into four groups randomly:Young control group, Old group, Zuogui Pill group and Yougui Pill group. The rats in Old group were administered D-Galactose 400mg · kg- 1 · d- 1 by intraperitoneal injection for 60 days consecutively and the rats in Young control group were administered the equal volume of normal sodium. And the intervention group were given Zuogui pill and Yougui Pill with 4.97g · kg-1 · d -1 or 4.94g · kg- 1 · d- 1 by intragastric administration at the same time respectively. The SOD activity and MDA contents in myocardium were measured. Western blot was used to detect the protein expressions of Bc1-2 and Bax and the Caspase - 3 relative activity in myocardium were detected too. Results :Compared with Young control group,the SOD activity was decreased and the content of MDA was increased in rat myocardium of the Old group. The protein expressions of Bcl - 2 were down regulated and reversely of the Bax. And the Caspase -3 relative activity was increased. The variations above in aging myoeardium were restored statistically in both of Zuogui pill group and Yougui Pill group. Conclusion : Zuogui pill and Yougui pill could slow down the heart aging progressthrough improving the anti-oxidation ability and decreasing the apoptosis.
