摘要
目的探讨丹参酮ⅡA对β淀粉样蛋白25-35片段(Aβ25-35)诱导的褐家鼠肾上腺嗜铬瘤细胞PC12细胞系抑制中的作用及机制。方法MTT检测丹参酮ⅡA(2μmol/L)对Aβ25-35(20mmol/L)处理过的细胞活性,应用公式(1-实验组平均OD值/对照组OD值)×100%计算细胞抑制率。RT-PCR、westernblot检测细胞内GSK3β及pi GSK3β的表达量。结果丹参酮ⅡA有效的拮抗了Aβ25-35引起的细胞抑制及GSK3β的高表达和低磷酸化。结论Aβ25-35可以引起PC12细胞抑制,而丹参酮可以拮抗Aβ25-35的作用,并且可能是通过GSK3β通路而发挥作用。
Objective To explore the role and the probabily mechanism of tanshineⅡA on β-amyloid25–35.Methods Application of MTT was used to detect the activity of PC12 cells.The formula(1-treatment group OD/control group OD) *100% was used to Calculate PC12 cells growth inhibition rate.Reverse transcription-PCR(RT-PCR) and western blot were used to observe the expression of GSK3β and piGSK3β.Results TanshineⅡA could effectively ease the cell inhibition induced by the Aβ25-35,the high expression of GSK3β and the low expression of piGSK3β.Conclusions Aβ25-35 could induce inhibition in PC12 cells and tanshineⅡA could ease the inhibition induced by the Aβ25-35 through GSK3 pathway.
