β淀粉样蛋白25-35片段最佳损伤建模条件的研究

摘要 为探讨淀粉样蛋白25-35片段(Αβ25-35)诱导细胞损伤的最佳条件,为后续Αβ作用机制研究奠定实验基础。采用正常血清培养和血清饥饿培养两种预处理方法,以Αβ25-35为诱导剂,建立SH-SY5Y细胞损伤模型,通过细胞形态学和存活率两个指标来确定损伤模型建立的最佳条件。结果显示,血清在Aβ25-35引起的SH-SY5Y细胞损伤中具有保护作用。由此可知,采用血清饥饿法培养SH-SY5Y细胞,以40μM的Aβ25-35处理细胞48h是建立细胞损伤模型的最佳条件。 To explore the optimal conditions forestablishing Aβ25- 35- induced SH- SY5 Y cell injury model, and to lay the foundation forthe mechanism study of Aβ thereafter. Serum- containing culture method and serum- free culture method were used in this study to establish the cell injury model. Morphological changes and cell viability were chosen to determine the best injury condition of Aβ25- 35. Ourresults showed that serum protects the cells from Aβ25- 35- induced injury; SH- SY5 Y cells treated with 40μM Aβ25- 35 in serum- free medium for48 h is the best condition forestablishing the Aβ25- 35- induced cell injury model.
