摘要
目的筛选能分离丹参总酚酸的高效、实用的树脂,并优化其吸附分离工艺。方法以丹酚酸B为指标成分,用紫外分光光度法测定总酚酸含量,考察树脂的吸附、解吸性能。结果HZ818大孔吸附树脂吸附和解吸性能较好,饱和吸附量为122 mg/g,上样最佳流速为2 BV/h(BV,床层体积),最佳解吸条件为5 BV 50%的乙醇溶液以2.4 BV/h流速进行洗脱,解吸率为93.8%。经HZ818分离得到的丹参总酚酸中丹酚酸B的纯度为65.7%,精制程度达到351.3%,回收率为90.5%。结论HZ818树脂性能优良,适合分离丹参总酚酸。
Objective To obtain an efficient macroporous resin for salvianolic acids absorption and desorption,and to optimize the method for salvianolic acids separation.Methods The concentration of salvianolic acid B was measured by UV spectrophotometry.The absorption and desorption properties of resins were studied.Results The absorption ability of HZ818 resin was 122 mg/g,the best loading flow rate was 2 BV/h(BV,bed volume),the best desorption rate was 93.8% with conditions of 5 BV of 50% ethanol as a elution and the desorption flow rate was 2.4 BV/h.The purity of salvianolic acid B was 65.7% with 351.3% purification factor and 90.5% recovery.Conclusion The resin of HZ818 was suitable for purification of salvianolic acids.
