摘要 将还原型辅酶Ⅰ(NADH)还原Fe3+生成Fe2+的化学反应与Fe2+催化溶解氧氧化Luminol的化学发光反应相偶合,同时采用试剂固定化技术将体系中的分析试剂Fe3+及Luminol全部电价固定在离子交换柱中,建立了一种测定还原型辅酶Ⅰ的流动注射化学发光新方法.该法对于NADH响应的线性范围为1.0×10-7~1.0×10-4mol/L,检出限为5.5×10-8mol/L,相对标准偏差小于5%(1.0×10-7mol/L,n=7). A novel chimluminescence(CL) based system for reduced coenzymeⅠ(NADH) combined with flow injection analysis is presented. The analytical reagents, luminol and Fe 3+ are immobilized on an anion exchange resin column and a cation exchange resin column, respectively. Fe 3+ is eluted by HCl solution passing through the cation exchenge column and mixes and reacts with the sample stream to generate Fe 2+ , which then merges with the luminol stream eluted by the injection of sodium phosphate through the anion exchange column under alkaline conditions to produce CL. The CL emission intensity is correlated with the standard NADH concentration in the range 1×10 -7 to 1×10 -4 mol/L, and the dection limit is 5.5×10 -8 mol/L. A complete analysis including sampling and washing could be performed in 1 min. The system is stable for 200 times with a relative standard deviation of less than 5%.
