可见光直接光度法测定硫代氧化型辅酶

摘要 建立了可见光直接光度法测定硫代氧化型辅酶。方法为通过确定测定试剂中各组分最佳含量,建立了一种运用3а-HSD偶联的酶法光度法测定Thio-NAD+。本法线性范围为0~2 mmol∕L,平均回收率为102.67%,批内变异系数(CV)和批间变异系数分别为1.50%~2.49%和2.44%~4.04%,线性回归方程和相关系数分别为Y=0.0467X+0.0033,r=0.9985。本法具有选择性好、灵敏度高、测定简便等特点,能真实反映Thio-NAD+浓度,对于提高Thio-NAD+检测的效率、结果的准确性都具有重要意义。 To establish a direct spectrophotometric method for the determination of the thiosulfate oxidized coenzyme (Thio - NAD). This 3a - HSD - coupled enzymatic assay for the analysis of Thio - NAD^+ was established by determinating each component as well as its content in the reagents. The method is liner in the range of 0 -2retool / L, the average rate of recovery is 103.00%, the withinrun and between -mn CV is 1.50% - 2.49% and 2.44% ~ 4.04%, the regression equations: Y =0. 0467X +0. 0033 ,r =0.9985. This method is selective, sensitive, easily measured, and truly reflect the Purity of Thio - NAD^+. There are of great significance for improving the efficiency of detection and the results of the accuracy.
