一株分离自竹黄产蒽醌类色素菌株的鉴定及其培养特性 被引量:3

Identification and Pigment-synthesis Capacity of a Producing Anthraquinone Pigment Strain Isolated from Shiraia bambusicola

摘要 以分离自竹黄菌天然子座的LBR-SB6菌株为出发菌株,通过对形态特征、培养特性的研究,确定了该菌株为曲霉菌属Aspergillus sp.以改良查氏培养基为基础,对LBR-SB6液态发酵碳氮源等条件作了初步探讨,结果表明:乳糖、麦芽糖和玉米浆、亚硝酸钠分别是菌株生长和产色素的最佳碳源和氮源,1.0%竹汁对菌株生物量和色素生成量具有明显的促进作用.最终确定了LBR-SB6菌株液态发酵培养的最佳条件为:乳糖2%,亚硝酸钠0.2%,玉米浆0.1%(以干物质计),1.0%竹汁,磷酸氢二钾0.1%,硫酸镁0.025%,氯化钾0.5%,硫酸亚铁0.001%;最适温度25-28℃,摇床转速150-200 r/min,起始pH值7.6-8.0.研究结果将为蒽醌类色素的生物合成以及在医药领域的研究应用提供一定的科学依据. A producing anthraquinone pigment strain LBR-SB6 was isolated from the stroma of Shiraia bambusicola.According to the characteristics of morphology and culture,the strain was identified as Aspergillus sp..Using modified-Czapek medium as culture,the liquid fermentation conditions of LBR-SB6 have been studied.Results show that the optimal carbon and nitrogen sources of LBR-SB6 are lactose,maltose,NaNO2,and corn steep liquor(CSL);that the effect of 1.0% bamboo juice on the strain's biomass and pigment-synthesis is significant;and that the optimal conditions in submerged culture with shake flask are: lactose 2%,NaNO2 0.2%,CSL 0.1%,bamboo juice 1.0%,K2HPO4 0.1%,KCl 0.5%,MgSO4·7H2O 0.025%,and FeSO4 0.001%;the most suitable temperature is 25~28℃,the rotation speed of shake flask is 150~200 r/min and the original pH value is 7.6~8.0.
