摘要
利用纤维素酶处理苹果渣提取多酚,通过单因素试验选取影响因素与水平,在单因素试验的基础上采用3因素3水平响应面分析法,对提取工艺进行优化设计,建立了纤维素酶法提取苹果渣多酚的数学模型,并对模型进行了验证。通过对二次回归方程的求解得到了最佳的酶解提取条件为:加酶量125 EGU/g、酶解温度50℃、酶解时间22 min,苹果渣多酚的提取量达到最大,最大值为3.433 mg/g。并且对酶法与传统水提取多酚的方法之间的比较做了初步研究,结果表明酶法比传统水提取方法提取多酚高27%。
To optimize the extraction condition of polyphenols from apple pomace treated with cellulase,the response surface methodology(RSM) was employed and three factors and three levels was designed based on the results of single factor tests.A quadratic equation for polyphenols extraction was derived and the effect of main factors and their crossproduct was studied,and finally the regression model was validated.By solving the quadratic regression equation using appropriate statistic methods, the optimal conditions of variables were determined as follows: enzyme amount 125 EGU/g, enzymolysis temperature 50 ℃, enzymolysis time 22 min. Under the conditions above, the yield of apple polyphenols was 3. 433 mg/g. The comparison of extraction efficiency between the methods treated with and without cellulose indicated that the extraction efficiency of total polyphenols could be improved by 27% through the treatment with cellulase.
