摘要
目的:探讨早孕因子(EPF)活性物对小鼠脾淋巴细胞增殖的影响及其在胎母免疫中的作用。方法:使用离子交换层析、亲和层析和凝胶过滤技术提纯3~6周孕妇血清中EPF;用淋巴细胞转化法观察EPF对淋巴细胞的作用。结果:EPF活性物G4、G3及孕血清均能抑制淋巴细胞DNA合成,尤以G4作用更为明显,并呈一定剂量依赖性。结论:EPF可能在介导、调控妊娠早期诱导的免疫抑制。
AIM:To investigate the effect of early pregnancy factor (EPF) on mouse splenocytes proliferation and its action on reproductive immunology.METHODS:EPF from the serum of pregnant women between 3~6 weeks of gestation was purified using ion-exchange chromatography,affinity chromatography and gelfiltration techniques.The effect of EPF on lymphocyte proliferation was observed with 3H-TdR incorporation technique.RESULTS:[WTBZ〗EPF-like substance (G4、G3)and pregnancy serum could inhibite lymphocyte DNA synthesis and G4 showed inhibitory effect more obviously which was does-dependent to some extent.CONCLUSION:EPF might have an important role in modulating maternal immunosuppression induced early by gestation and ensuring the continuation of pregnancy.
