摘要
用改良的Mehta方法,以从混合孕妇(5~12周)血清中提取出的早孕因子作对照,对混合恶葡、绒癌患者血清中早孕因子样物质(t—EPF)进行分离纯化,测定各阶段提取物中EPF活性、hCG—β及蛋白质含量,并经PAGE及SDS—PAGE鉴定提取效果、测定肿瘤源性早孕因子的分子量。获得不含hCG具EPF活性的t—EPF(TG_3. TG_4)提取物,TG_3中两条多肽成份分子量为27.78ku及57.046ku,TG_4电泳显示单一多肽区带。由TG_3中两条多肽的分子量及其近倍数关系推测恶性滋养叶细胞肿瘤患者血清中的t—EPF可能不同于孕血清中的EPF,且t—EPF可能以单体及聚合体形式存在,其具活性的基本形式为27.78ku的多肽。
Early pregnancy factor (EPF)—like substance in the pooled sera of patients with malignant hydatidiform mole and choriocarcinoma was isolated and purirified by the modified Mehta's method, with EPF extracted from the plooled sera of early pregnant women (5~12 weeks) as control, EPF activity, hCG—β and protein contents were determined at various stages. The effect of purification and the molecular weight of t—EPF were detected by polyacrylamide gel electrophoresis (PAGE) and SDS—PAGEtechniques. The extracts of t—EPF(TG_3, TG_4) which were found to be positive for EPF activity and negative for hCG—β activity were obtained from the pooled sera of patients with malignant hydatidiform mole and choriocarcinoma. Two seperate polypeptides bands in TG_3 were showed by PAGE and their molecular weights were 28 and 57.5 KD by SDS—PAGE. Single polypeptide band in TG_4 was shown by PAGE. From the molecular weights of two seperate polypeptides and their multiple relationship, we presumed that t—EPF present in the sera of patients with malignant trophoblastic tumors might differ from EPF present in the pregnant serum and, it could exist in the states of monomer and polymer. The basic active form of t—EPF is a polypeptide of 28 KD.
