摘要
目的研究姜黄素对人肺癌细胞诱导凋亡的作用机制。方法用10、20μM两种浓度姜黄素分别作用于SPC-A1细胞12h和24h,用末端标记法检测细胞凋亡率,原位杂交检测姜黄素作用24h后SPC-A1细胞凋亡相关蛋白Survivin表达。结果20μM姜黄素作用24h组凋亡率达43.7%,明显高于其他组;20μM姜黄素作用24h组凋亡相关蛋白Survivin mRNA表达明显降低。结论姜黄素可诱导人肺癌细胞SPC-A1凋亡,其作用机制可能与凋亡相关蛋白Survivin mRNA表达降低有关。
Objection To investigate the mechanism of anti-tumor effects of cureumin on human lung cancer cell SPC-A1. Methotis SPC-A1 cells were exposed to cureumin at different concentration of 10,20 p,M. Apoptotic index was determined by in situ end-labe-ling method. In situ hybridization was employed to determine Survivin expression. Results The TUNEL method certified that apoptotic ceils reached 43.67% at the concentration of 20μM curcumin at 24h. There are significant difference between cucurmin groups and control group. Treated with 20 μM eurcumin for 24 h, the expression of Survivin mRNA of human SPC-A1 cells was down-regulated. Conclusions Curcumin can induce the apoptosis of SPC-A1 cells, and one of its molecular mechanism may be associated with the down-regula-ting expression of Survivin.
