摘要
目的 研究骨髓增生异常综合征 ( MDS)细胞株 MU TZ- 1凋亡相关基因 Survivin、bcl- 2和 bax的表达及高三尖杉酯碱 ( HHT)诱导其凋亡的作用机制。方法 用 MDS- RAEB细胞系 MU TZ- 1为体外模型 ,采用透射电镜和流式细胞仪 Annexin- VF ITC/ PI双染分析细胞凋亡 ,PI染色分析细胞周期变化 ,RT- PCR技术检测抗凋亡基因Survivin、bcl- 2和促凋亡基因 bax的表达。结果 HHT能诱导 MU TZ- 1细胞凋亡 ,凋亡率与作用浓度和时间成正相关 ,细胞被阻滞在 G2 期。HHT作用 MU TZ- 1细胞 6小时后细胞内抗凋亡基因 Survivin表达明显下降 ,而抗凋亡基因 bcl- 2和促凋亡基因 bax表达无明显变化。结论 HHT能诱导 MUTZ- 1细胞凋亡 ,抗凋亡基因 Survivinm RNA下调可能是 MU TZ-
Objective To investigate the expression of apoptosis related genes, Survivin, bcl 2 and bax in apoptosis of MDS cell line MUTZ 1, and possible mechanisms of homoharringtonine (HHT) in the treatment of myelodysplastic syndrome (MDS). Methods The apoptosis of MDS RAEB cell line MUTZ 1 was observed by transmission electron microscope. Cell apoptosis and cell cycle shift were also investigated by flow cytometry (FCM).The expression of Survivin, bcl 2 and bax was detected by retrotranscriptase polymerase chain reaction (RT PCR). Results Apoptotic peak on FCM and positive Annexin V FITC on cell membrane showed that HHT induced apoptosis of MUTZ 1 cell in a dose and time dependent manner. HHT significantly down regulated Survivin expression, but there were no changes on bax and bcl 2 expression. Conclusion HHT could induce apoptosis of MDS cell line(MTUZ 1) and Survivin might play an important role in MDS.
