生长激素对脐血来源树突细胞成熟状态的影响

Growth hormone influence on functional status of cord blood-derived dendritic cells

摘要 目的检测生长激素(GH)对脐血来源树突细胞(DC)的免疫表型和功能状态的影响。方法从新鲜脐带血中分离出单核细胞,应用白细胞介素4(IL-4)和重组人粒一巨细胞集落刺激因子(rhGM—CSF)诱导刺激单核细胞,以不同浓度的GH作为修饰因子修饰DC诱导过程;观察DC形态、数目、生长状态,流式细胞仪检测DC的免疫表型,酶联免疫吸附试验测定DC上清液中自细胞介素12(IL-12),自体混合淋巴细胞反应测定其功能状态.结果脐血贴壁的单核细胞在细胞因子和GH诱导下可以分化为成熟DC,不同浓度(5、25、100ng/ml)GH可显著上调DC CD83(31%±4%、34%±4%、34%±5%)、CD80(26%±4%、29%±6%、30%±6%)和HLA—DR(45%±8%、49%±6%、50%±5%)分子表达的阳性率(P〈0.05),明显提高DC分泌IL-12的能力[分别由第5天的(91±12)、(159±20)、(166±19)pg/ml上升到第9天的(686±58)、(796±61)、(810±67)pg/ml](P〈0.05),DC对自体淋巴细胞的激活功能明显增强(P〈0.05)。结论GH可明显促进DC的诱导过程,修饰加速DC的成熟并提高细胞表面共刺激分子表达,增强DCIL-12分泌和激发自身淋巴细胞增殖能力;GH的修饰作用与浓度有一定的相关性。 Objective To detect the effect of functional status and immunophenotype of dendritic cell modified with growth hormone (GH) from cord blood. Methods Monocyte was isolated from fresh cord blood. DCs were induced with IL-4 & GM-CSF and modified with different concentrations of GH. The morphology, count and growth status of DC were observed appearance. And the immunophenotype of DC was detected with flow cytometry. IL-12 in supernatant was measured with ELISA. The immuno-activity was determined with autologous mixed lymphocyte reaction. Results Mature DC were induced from adherent lymphomonocyte under the action of IL-4, GM-CSF and GH of different densities (5, 25, 100 ng/ml). GH increased the positive expression of CD83 (31% ± 4% , 34% ± 4% , 34% ± 5% ), CD80 (26% ± 4% , 29% ±6%, 30% ±6% )and HLA-DR(45% ±8%, 49% ±6%, 50% ±5% ) (P 〈0. 05), enhanced the ability of secreting IL-12 ( P 〈 0. 05 ) and boosted significantly the immuno-activation of DC to autologous lymphocyte (P 〈 0. 05). Conclusion GH could promote the differentiation, maturation and expression of co- stimulatory molecules, increase the IL-12 production in DC and enhance the ability of DC to stimulate the proliferation of lymphocyte within a certain dose range.
