摘要
目的研究重组人生长激素(rHGH)对人直肠癌小鼠肾包膜下移植瘤生长的影响。方法雄性BALB/C小鼠45只术前24h一次性皮下注射环磷酰胺(CTX)100mg/kg,然后行人直肠癌小鼠肾被膜下移植术(SRCA),随机分成rhGH1组、rhGH2组每日皮下注射rhGH3IU/kg,联合组、化疗组和对照组连续用药7d。各组小鼠随机留3只待其自然死亡,观察其生存期,余小鼠术后10d处死,观察小鼠外周血细胞数、体重、移植瘤末初体积差(△TS)、抑瘤率(IR)及(HE染色)组织学变化。结果联合组及化疗组移植瘤△TS及IR比前三组差异有显著性(P<0.01);联合组和化疗组平均生存期显著高于对照组(P<0.05);联合组白细胞数值显著高于化疗组(P<0.05);对照组和rhGH各组的组织病理形态相似,而联合组和化疗组表现为实质细胞减少,间质增多。结论rhGH对人直肠癌小鼠肾包膜下移植瘤生长无明显促进作用;rhGH和化疗药联用对化疗药的抑瘤作用无明显影响、可减轻化疗药的骨髓抑制反应;二者联用可延长实验小鼠的生存期。
Objective To investigate the effect of rhGH on the growth of rectal cancer in SRCA. Methods Forty five male mice BALB/C subcutaneously injected with CTX 100mg/kg per mouse before operation, were randomly divided into five groups. SRCA was carried out simultaneously with fresh explants of human careinoma of rectum in all animals. From the first to the seventh day after surgery, rhGH1 group received i.h. rhGH 1IU/kg daily, rhGH2 group received i.h. rhGH 3IU/kg daily, rhGH + 5 - FU group received i.h. rhGH 1IU/kg daily and i.e 5 - FU 20mg/kg, 5 - FU group received i.c. 5 - FU 20mg/kg daily. While control group daily received i.h. 0.2ml and i.c. 0.5ml sterile saline. The body weights of the animals were measured every other day. On the lOth day after surgery, 6 mice in each group were executed to observe the blood routines, △TS, IR and the differences of HE dyeing, while the others were fed till death to obverse survival time. Results The △TS and IR of rhGH 1 + 5 - FU group and 5 - FU group distinctly decreased as compared with that of control group, rhGH1 group and rhGH2 group (P〈0.01 ) . The lifespan of animals in rhGH1 + 5 - FU group and 5 - FU group significantly increased as compared with that of NS group (P〈0.05 ). The number of white cells of rhGH1+ 5 - FU group remarkably increased compared with 5 - FU group ( P〈0.05) . Compared with that of rhGHl group and rhGH2 group more interstitial tissue and less parenchyma cells were noticed in rhGHl + 5 - FU group and 5 - FU group. Conclusion In vivo rhGH does not obviously stimulate the growth of the transplanted tumor of rectal cancer in SRCA. The inhibited effect of 5 - FU on the transplanted tumor could not be signiflcantlyaffected by rhGH. The rhGH combined with 5 - FU can offset the adverse effect of 5 - FU on the blood of animals and elongate the lifespan of tumor bearing mice.
