摘要
本文采用DNA凝胶电脉、TUNEL及超微结构观察等技术,观察了常氧和模拟海拨7000米低氧2、4、6、8d大鼠脑组织变化。低氧6、8d脑组织凝胶电泳出现梯状带,其它各组没见上述变化。TUNEL法观察到程序性死亡的细胞染成棕黄色,其它细胞不着色,并且着色细胞分布有区域性,主要分布在皮层、海马和丘脑。超微结构观察到一些细胞出现程序性死亡的特征,胞浆空泡化,染色质凝集,边缘化,形成凋亡小体等变化。上述结果表明,低氧能导致大鼠脑细胞的程序性死亡,或程序性死亡至少是低氧性脑损伤的一个因素。

In this study,agrosegel electrophoresis,Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling and ultrastructure examination were used to observe normal and hypoxic rats brain cells programmed cell death(PCD) or apoptosis(AP).Hypoxic rats were put into hypobaric chamber at a simulated altitude of 7000m for 5h each day during 2、4、6、8 days hypoxic exposure.During hypoxia,the brain cells degenerated and expressed apoptotic characteristics.Ultrastructure examination showed the condensation of both the nucleus and cytoplasm following 2 days hypoxia,then fragments and pinches off at the surface protuberances.Finally,apoptotic bodies were observed.TUNEL stain showed that the apoptotic cells were stained brownyellow,and expressed regional nature.It most likely emerged in cerebral cortex,thalamus and hippocampus.Positive TUNELstain was found in cerebral cortex as early as 2 days hypoxia,then in hippocampus and thalamus after 4 days hypoxia.But the ladderlike pattern of DNA was not observed until 6 or 8 dyas hypoxia.This study suggested that hypobaric hypoxia can induce brain cells apoptosis or that an apoptotic mechanism accounts for at least one component of brain damage following high altitude hypoxia.
