摘要 采用“搓网”方法将8种96例不同的恶性肿瘤手术切除标本制成细胞膜完整的单细胞悬液,并从6例癌性腹水中提取富含癌细胞的悬液。用流式细胞免疫学方法检测癌细胞(MDR1)表达产物P170含量。经上述方法制成悬液中的癌细胞丰富、荧光显微镜下观察胞膜完整,P170阳性细胞抗体荧光位于胞膜上。本文认为用流式细胞免疫学技术检测恶性实体瘤细胞MDR1表达产物P170含量简便易行,不仅可以了解肿瘤细胞始基的药物敏感性,还可在化疗过程中动态监测药物敏感性的变化。 To explore the feasibility of FCM in studying the multidrug resistant gene (MDR1) expression in malignant solid tumors,singlecell suspension with intact cellular membrane taken from 96 operative specimens of 8 different kinds of malignant solid tumor were prepared using rubbingnet technique.Autologous tumor cell (ATC) suspension was also made from 6 ascites samples of advanced ovarian carcinomas. The suspensions were assayed for their MDR1 product P170 (Pgp) contents by FCM immunology. It was found that the suspensions were rich in single neoplastic cells with intact cell membrane. The labelled fluorescence of P170positive cells spotted the membrane. This exemplifies that this technic is highly feasible in studying MDR1 expression of malignant solid tumors and is helpful to perceive inherent nature of multidrug resistance.
