摘要
目的建立反相高效液相色谱(RP—HPLC)法测定苦丁茶含片中槲皮素和山柰素的含量。方法以Nova-Pak C18(3.9mm×300mm,4.0μm)为色谱柱;甲醇-0.4%磷酸溶液(50:50)为流动相;检测波长360nm;流速0.7ml·min^-1;柱温40℃。结果槲皮素的进样量在0.01299~0.6496μg范围内,山柰素的进样量在0.01334—0.6672μg范围内与峰面积呈良好的线性关系。平均加样回收率分别为97.03%和97.34%,RSD分别为1.16%(n=6)及1.42%(n=6)。结论该方法简便、快速、准确,可作为苦丁茶含片中槲皮素和山柰素的含量测定方法。
Objective To establish an RP - HPLC method for determination of the content of quertetin and kaempferol in Kudindcha tablets. Methods The Nova -Pak C18 column (3.9 mm× 300 mm,4. 0μm) was used. The mobile phase was methanol - 0.4% H3 PO4 (50: 50), the detection wavelength was 360nm, the flow rate was 0. 7 ml·min^-1, the temperature was 40℃. Results The linear range of quertetin was 0.012 99 ~0. 649 6μg , the linear range of kaempferol was 0.013 34 ~0.667 2μg. The average recovery was 97.03% and 97.34% ,RSD was 1.16% (n = 6) and 1.42% (n = 6). Conclusion This method is simple, rapid and accurate. It can be used for determination of the content of quertetin and kaempferol in Kudindcha tablets.
