摘要
研究了黑曲霉Aspergillus niger ZJUT712菌株固态发酵和水煮工艺相耦合炮制牛蒡子.结果表明,该工艺提高了牛蒡子中有效成分牛蒡子苷元的含量,从而有利于促进牛蒡子摄入体内迅速起效.最佳固态发酵炮制条件为:0.5g牛蒡子粉、3g麸皮、2g甘蔗渣、0.3g蛋白胨和10ml Mandels营养液,固液比1∶3.6g/ml,初始pH5.6,30℃发酵7d.牛蒡子苷元产率随底物初始浓度的增加而降低.A.nigerZJUT712转化0.174mmol/L牛蒡子苷的产率可达93.0%.7d时间内A.niger ZJUT712水解牛蒡子苷的过程符合Monod方程,反应动力学常数为Vm=0.0837mmol/(L.d),Km=0.16mmol/L.
The processing of fructus arctii, a Chinese medicine, by solid-state fermentation using Aspergillus niger ZJUT712 coupled with poach has been studied. The results show that this technique can significantly enhance the arctigenin content in fructus arctii. The optimum medium components are 0.5 g fruetus arctii powder, 3 g bran, 2 g bagasse, 0.3 g peptone, and 10 ml Mandels nutrient fluid. Solid-to-liquid ratio is 1:3.6 g/ml. Initial pH of the medium is 5.6. Lower yield was observed with higher initial arctiin concentration used. The yield reached 93.0% when 0.174 mmol/L arctiin was hydrolyzed at 30 ℃ for 7 d. In 7 d, the hydrolysis of arctiin is in accordance with the equation of Monod. The kinetic constants are Vm=0.083 7 mmol/(L·d) and Km= 0.16 mmol/L.
