摘要
本文探讨了在非水相介质中酶催化逆水解反应合成红景天甙的新方法,研究了非水相反应体系、酪醇浓度、D-葡萄糖浓度、反应时间、pH值对β-葡萄糖苷酶催化酪醇和D-葡萄糖合成红景天甙的影响,优化了酶法合成红景天甙的条件,使其转化率达到了17.7%,即7.39 g/L,远远高于米曲霉整体细胞催化的产量(0.7 g/L)。该方法可望应用于其它具有生理活性糖苷类化合物的高效酶促合成,具有潜在的应用价值。
This paper studied the enzymatic synthesis of salidroside with tyrosol and D-glucose in monophasic aqueousorganic medium, using a crude β-D-glucosidase from Aspergillus oryzae as a catalyst. The effects of nonaqueous reaction system ,concentration of D-glucose and tyrosol, reaction time and pH on the synthesis of salidroside were investigated. The salidroside yield reached 7.39 g/L under the optimum conditions,which was much higher than the yield of 0.7 g/ L catalyzed by the whole ceils of Aspergillus oryzae. A monophasic organic-water medium system for efficient enzymatic synthesis of salidroside by reversed hydrolysis has been constructed and optimized. It is expected that this organic solvents-buffer system can be applied to the synthesis of other physiologically active glucosides.
