摘要
目的:研究两面针结晶8的镇痛作用并对其镇痛机制进行初步探究。方法:①采用热板法、扭体法观察结晶8的镇痛效果;②采用HPLC测定小鼠血清及脑组织中的单胺类递质的含量变化。结果:①结晶8高低剂量组均对热板法所至小鼠的疼痛有明显的抑制效果,能使小鼠血清中的5-T含量减少(P<0.05或0.01);②结晶8低剂量组对小鼠的扭体反应有明显的抑制效果,能使小鼠脑内的DA含量减少(P<0.05)。结论:结晶8具有镇痛作用,而镇痛机制可能与其降低脑内DA和外周组织5-HT含量有关。
Objective:To investigate the analgesic effect of the crystal 8 from Zanthoxylum nitidum(Roxh. DC. and study its analgesic mechanism. Methods:(1)The analgesic effect of Crystal 8 was respectively evalu- ated by the writhing test and hot-plate test in mice. (2)The changes of the Monoamine neurotransmitter contents in the serum and brain of the mice were observed by HPLC. Result: (1)In the high dosage group and the low dosage group, the pain threshold value of the mice was enhanced obviously in the hot-plate test and the contents of 5-HT was reduced in their serum. ( P 〈0.05 or 0.01). (2)In the low dosage group, the writhing times of the mice was reduced markedly and the contents of DA was reduced in the brain of the mice ( P 〈0.05). Conclusion:The Crystal 8 has markedly analgesic effect and its analgesic mechanism may be related to reduction of the contents of DA in the brain and 5-HT in the peripheral tissue.
