摘要
采用以羟磷灰石和α-磷酸三钙两晶相为基的无机骨水泥(HAC)作为药物缓释载体,载入约8%去甲万古霉素(NVCM)药物后组成复合体,用于动物实验性骨缺损合并骨感染伤口的治疗,探讨HAC载药物缓释系统在骨感染预防方面的临床应用价值。通过局部用药、全身用药和不用药等不同方法对骨感染伤口进行处理,根据动物术后体温、体重、血常规、全身和局部反应状况以及组织学检查等指标来观察、比较其局部骨感染的差异。结果表明,骨感染后及时用药可有效地控制局部炎症的进一步发展,单纯HAC并没有抗感染的功能;全身用药比局部用药更能抑制或延缓全身反应的发生;载药复合体虽然未能完全消除感染,但可以显著延缓感染的迅速发展。
To investigate the competence of hydroxyapatite bone cement with 8% norvancomycin (HAC/NVCM composite) as an implantable drug delivery system for prophylaxis of bone infection. The bone defects and infection models in rabbits were first established. Then the animals were divided into four groups:Group A with the HAC/NVCM composite putting in the defect;Group B were used NVCM solution injected through rabbits' ear' s vein with no drug loaded HAC putting in;group C haven't use any antibiotics with no drug loaded HAC putting in; group D with no antibiotics and no drug loaded HAC putting in. The differences of bone infection and the wound healing were compare by body weight, body temperature, routine blood count and histological observation. The results indicated that HAC/NVCM composite can effectively control the inflammation reaction and promote the wound healing. It can widely be used in the prophylaxis and treatment of bone infection and bone deject.
