摘要
对一株能粗降解铬革屑菌株的发酵条件进行优化,通过Folin试剂法测定了不同发酵条件下发酵液的蛋白酶活性,确定了菌株W_(1-1)的最适产酶条件。研究表明:菌株的最适产酶条件为葡萄糖9.0%,酵母膏0.5%,CaCl_20.3%,K_2HPO_40.3%,MgSO_4 0.2%,初始pH 8.5,装液量90mL(90/250mL三角瓶),接种量4.0%,种龄14h,发酵温度37℃,发酵时间36h。在此条件下,测得发酵液的蛋白酶活力为37.36u/mL,比优化前的产酶量提高了35.4%;最后,采用该菌株在最适产酶条件下对铬革屑进行降解,降解率达到71.38%。
The fermentation conditions of a strains for degradation chrome shavings was optimized. After shaking flask culture, the protease activity on different fermentation conditions by Folin reagent was tested, and the optimum conditions of producting protease was obtained as follows : glucose 9.0%, yeast extract 0.5% , CaCl2 0.3% , K2HPO4 0.3% , MgSO4 0.2% , initial pH 8.5, loading liquid weight 90mL(90/250mL triangular flask), inoculum 4%, age 14h, 37℃, fermentation 36 h. On such conditions, the protease activity of strains was 37.36u/mL, increasing by 35.4% ,and degradation rate of chrome shavings was 71.38%.
