摘要 比较了长江华溪蟹Sinopotamon yangtsekiense心脏、肝胰腺、鳃和性腺等组织中提取总RNA的4种方法,同时比较了同一方法提取不同组织RNA的质量,总结出适合于蟹类组织RNA提取的方法。结果表明,TRIzol法提取效果最好,心脏总RNA质量最高。此外,实验中提取的总RNA污染少、完整性好,为以后进行cDNA合成、基因表达及定量分析等研究奠定了基础。 The technique of RNA isolation is frequently used in modem biology research. Different methods were compared to obtain total RNA from the tissues of freshwater crab Sinopotamon yangtsekiense, such as heart, hepatopancreas, gill and gonad. RNA was extracted with the same method from different tissues of the sample. A method for crab RNA isolation was summarized. The results showed that TRIzol method was the best among the four methods, and the quality of RNA from the heart was the highest among the four tissues. The extracted total RNA was intact and without contamination, and could be used be for cDNA (complement DNA) synthesis and quantitive study of gene expression based on RNA isolation.
