摘要
目的:检测肺腺癌细胞内顺铂浓度,从而研究肺腺癌对顺铂的耐药与细胞内顺铂浓度的关系。方法:用四甲基偶氮唑蓝(methylthiazolyl tetrazolium,MTT)法测定 A549/DDP 细胞的耐药倍数,用 ICP-MS 测定细胞内顺铂(diammine dichloroplati-num,DDP)含量。结果:铂浓度在0.5~50μg·L^(-1)范围内线性关系良好,铂最低检出限为0.029μg·L^(-1),RSD 为0.41%~4.7%,回收率为98.2%~99.7%。耐药细胞 A549/DDP 内顺铂的浓度显著低于 A549细胞。结论:首次建立了 ICP-MS 测定细胞内顺铂浓度的方法。本方法准确、灵敏、简便,可用于测定细胞内顺铂浓度及肿瘤细胞耐顺铂的研究。
Objective: Determination of intracellular cisplatin in human lung ladenocarcinoma cell and study the relationship of drug - resistance and its content. Methods :The drug - resistance of A549/DDP to DDP was tested by MTF assay. The content of intracellular cisplatin in cell was tested by inductively coupled plasma mass spectrometry ( ICP - MS). Results :The calibration curve was linear when the concent of the Pt were in the range of 0. 5 - 50 μg·L^-1(r =0. 9997). The detection limit of Pt was 0. 029 μg·L^-1. The average rate of recovery of Pt was 98.2% -99.7% , with RSD 0.41% -4.7% (n = 5 ). The accumulation of DDP in A549/DDP were remarkably lower than these in A549 cell. Conclusion : ICP - MS determination of the content of intracellular cisplatin in human lung ladenocarcinoma cell is established for the first time. The method is accurate, sensitive and simple. It can be used for the determination of the content of intracellular cisplatin and the study of resistance of human lung ladenocarcinoma cell to cisplatin.
