摘要
[目的]探讨纤维素酶的最佳提取工艺和最佳反应条件。[方法]以里氏木霉Rut C-30为发酵菌种,在30℃、摇床转速170 r/min条件下培养8 d,发酵生产纤维素酶,用盐析技术对粗酶液进行分离纯化,通过正交实验法探讨了纤维素酶的提取工艺条件。并以羧甲基纤维素钠酶活力为指标,对该酶的最佳反应条件和稳定性进行了研究。[结果]纤维素酶的最佳提取条件是:提取时间为16 h、盐析饱和度为70%、pH值为4.8。纤维素酶的最佳反应条件是:pH值为4.8、温度为60℃。酶在pH 3.6~7.0时较稳定,在78℃保温30 min下的残留酶活为50%。[结论]该研究为酶的工业化生产提供参考数据。
[Objective] The aim of the study was to discuss the optimum extraction technology and reaction condition of cellulase.[Method] With Trichaderma reesei Rut C-30 as fermentation strain,it was cultured at 30 ℃ and rotation speed of 170 r/min for 8 d to produce cellulase by fermentation.The crude enzyme solution was purified by salting-out technology.The technological condition of extracting cellulase was discussed through orthogonal experiment.And with the activity of CMC-Na as index,the optimum reaction condition and stability of the cellulase were studied.[Result] The optimum extraction condition of cellulase was extraction time of 16 h,salting-out degree of 70% and pH value of 4.8.The optimum reaction condition of cellulase was pH value of 4.8 and temperature of 60 ℃.When pH value was 3.6-7.0,the cellulase was relatively stable,its residual enzyme activity was 50% after it was kept at 78 ℃ for 30 min.[Conclusion] The study provided reference data for the industrial production of enzyme.
