摘要
目的探讨茶黄素对大鼠局灶性脑缺血再灌注损伤后细胞凋亡及凋亡相关基因的影响。方法采用线拴法建立大鼠大脑中动脉栓塞(MCAO)模型。大鼠随机分为缺血组,假手术组,茶黄素小剂量组(10 mg/kg),茶黄素大剂量组(20mg/kg)。缺血2h,再灌注24 h后处死动物,于再灌注前舌下静脉给药。测定大鼠神经系统症状,脑梗塞体积,脑组织MDA,NO含量以及SOD和GSH-Px活性,采用TUNEL法检测脑组织细胞凋亡,采用免疫组织化学法检测凋亡相关基因Caspase-3的表达。结果茶黄素治疗组与模型组相比神经系统症状减轻,脑梗塞体积缩小,脑组织MDA,NO含量减少,SOD和GSH-Px活性增加。脑组织细胞凋亡及Caspase-3表达减少。结论茶黄素对大鼠局灶性脑缺血再灌注损伤具有保护作用,其作用机制可能与抗氧化和抑制细胞凋亡有关。
Objective To study the effect of theaflavin on cell apoptosis and apoptosis related gene after focal cerebral ischemia injury in a rat model of middle cerebral artery occlusion (MCAO). Methods Male Sprague - Dawley rats were anesthetized and subjected to 2 h of MCAO followed by 24 h reperfusion. The neurological deficits and the sizes of cerebral infarction were measured. The contents of malondialdehyde (MDA), and nitric oxide (NO) in brains were tested. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH - Px) were also tested. TUNEL staining to label the neuronal apoptosis and immunohistochenfistry to detect the expression of caspase - 3 protein were used. Results The neurological deficits and the sizes of cerebral infarction were decreased in theafiavin - treated groups. The contents of MDA and NO in theafiavin - treated groups were lower than those of the ischemic group. The activities of SOD and GSH - Px in theaflavin - treated groups were higher than that of the ischemic group. Theaflavin - treatment also decreased neuronal apoptosis and the expression of caspase - 3 protein. Conclusion Theaflavin has protective effects on focal cerebral ischemic reperfusion injury through inhibiting oxidative stress and lessening apoptosis.
