摘要 目的检测CD59位点短肽封条对HeLa细胞凋亡相关基因caspase-3、survivin表达水平的影响,探讨CD59位点短肽封条促肿瘤细胞凋亡的可能机制。方法将CD59位点短肽封条作用于HeLa细胞,利用免疫组织化学方法检测短肽封条作用12 h和24 h后HeLa细胞caspase-3、survivin的表达水平。结果caspase-3表达随短肽封条剂量和作用时间的增加而增加;survivin表达随短肽封条的剂量和作用时间的增加而减少。结论CD59位点短肽封条能下调survivin的表达并活化caspase-3,从而可能导致HeLa细胞的凋亡。 Objective To study the effect of peptide seals on CD59 to HeLa cells, detect expression of apoptosis-related gene survivin, caspase-3 in HeLa cell and investigate the mechanism of peptide seals to CD59 in inducing apoptosis of HeLa cells. Methods The peptide seals to CD59 were put into HeLa cells. Expressions of caspase-3 and survivin were detected by immunohistochemistry after interaction with peptide seals for 12 and 24 hours, respectively. Results Expression of caspase-3 increased, while survivin decreased with increase of dosage of the peptide seals and prolongation of action time. Conclusion The peptide seal to CD59 can down-regulate the expression of survivin, activate caspase-3 and play a great role in enhancing the apoptosis of HeLa cells.
