摘要
目的分析雌激素受体α(ERα)基因XbaⅠ和PvuⅡ酶切多态性对系统性红斑狼疮(SLE)的遗传影响。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对70例女性SLE患者和200名正常女性对照者的ERα基因(XbaⅠ,PvuⅡ位点)进行分型,并计算基因分布频率。结果女性SLE患者的ERα基因多态性分布中X和P的基因频率与正常对照组并无显著性差异,ERα基因Pp基因亚型的阳性率高于正常对照组(P(0.05)。SLE患者的ppXx基因型的阳性率明显高于正常对照组,而ppxx基因型的阳性率明显低于正常对照组(P(0.05)。结论女性SLE患者ERα基因的X和P的基因频率表达并无明显增高,但ERα基因酶切多态性分析中,Pp及ppXx基因型可能与女性SLE的易感性有关。
Objective To analyze the genetic influence of estrogen receptor alpha (ERα) gene polymorphism in female patients with systemic lupus erythematosus (SLE). Methods Xbal and Pvu Ⅱ in ERα gene in 70 female patients with SLE and 200 female healthy controls were typed by PCR-RFLP. Results There was no significant difference for X and P allele frequencies between SLE group and healthy control group ( P 〉 0.05 ). The frequency of Pp genotype of ERα gene was higher in female SLE patients than that in healthy control ( P 〈 0.05 ). The frequency of ppXx genotype of ERα gene in female SLE patients was significantly increased, while the frequency of ppxx genotype was significantly decreased ( P 〉 0.05 ). Conclusion The frequencies of X and P genotype were not significantly increased in Chinese female patients with SLE, but the Pp and ppXx genotype of ERα gene may be associated with the female SLE patients in analysis of restriction enzyme digestion.
