摘要
目的探讨谷氨酸和γ-氨基丁酸对海马神经干细胞诱导分化的影响。方法取新生Wistar小鼠海马组织,进行体外培养,分成5μmol/L(A组)、25μmol/L(B组)浓度的谷氨酸和5μmol/L(C组)、25μmol/L(D组)浓度的γ-氨基丁酸4个组及空白对照组(E组),分别对神经干细胞进行诱导分化,于分化的第7和14天用免疫荧光染色方法观察神经干细胞分化成神经元和神经胶质细胞的比例。结果在分化的第7天,免疫荧光显示,A、B、C、D组神经元比例即微管相关蛋白2ab(MAP2ab)阳性率均高于E组(P<0.05)。且B、D两组神经元比例即MAP2ab阳性率均分别高于A、C组(P<0.05)。而分化的第14天,神经元比例明显降低,但仍高于E组(P<0.05),而神经胶质细胞增多,且A、C两组神经元比例即MAP2ab阳性率均分别高于B、D组(P<0.05)。结论谷氨酸、γ-氨基丁酸均促进神经干细胞向神经元方向分化,但最佳浓度可能随分化时间的不同而进行调整。
Objective To investigate the effect of glutamate and γ-aminobutyric acid on differentiation of hippocampal stem cells.Methods Neonatal rat hippocampal stem cells were isolated and cultured in vitro.Then the differentiation of hippocampal stem cells were induced by 5 μmol/L(group A),25 μmol/L(group B)glutamate and 5 μmol/L(group C),25 μmol/L(group D)γ-aminobutyric acid respectively.The hippocampal stem cells without interference were chosen as control group(E).At day 7 and day 14 after differentiation,the ratios of neurons and glial cells were determined with immunofluorescence.Results At day 7 after differentiation,the ratios of the neural cells in groups A,B,C,D were higher than in group E(P〈0.05).And the ratios of the neural cells in groups B and D were higher than in groups A and C,respectively(P〈0.05).At day 14 after differentiation,the ratio of neuron cells in each group decreased significantly compared with that at day 7 after differentiation,but it was still higher than in group E(P〈0.05).and the ratios of the neural cells in groups A and C were higher than in groups B and D(P〈0.05).While the ratio of glial cells increased in each group at day 14 after differentiation compared with that at day 7.Conclusion Both glutamate and aminobutyric acid can induce the hippocampal neural stem cells to differentiate into neurons,but the optimal concentration should be adjusted at different times.
