摘要
目的:通过观察不同的细胞外基质对神经干细胞(NSCs)分化的影响,寻找促使NSCs向神经元分化的最佳细胞外基质,为NSCs定向分化培养及NSCs共移植体的构建提供实验依据。方法:利用无血清培养和细胞克隆培养技术,从新生大鼠海马分离NSCs,进行体外扩增、悬浮培养、传代;将层粘连蛋白(LN)、Matrigel、多聚赖氨酸与NSCs贴壁混合培养14d;采用免疫细胞化学和免疫荧光技术,观察NSCs的分化情况。结果:通过上述实验表明,在利用不同的细胞外基质混合培养时,LN组MAP2阳性细胞数明显高于其它两组;多聚赖氨酸组和Matrigel组GFAP阳性细胞数明显高于LN组。结论:LN有助于NSCs向神经元定向分化,是NSCs共移植体较理想的细胞外基质。
Objective:To find the best extracellular matrix which promotes the differentiation of neural stem cells to neurons by studying the effects of different extracellular matrix on differentiation of neural stem ceils so as to provide experimental evidence for the neural stem cell culture orientation- differentiation and the construction of the co-transplanted implant of neural stem ceils. Methods: The serum free and clone culturing technology was used to isolate, culture and hand down neural stem cells from neonatal rat. Poly-1-lysine,Laminin(LN) and matrigel were co-cultured with neural stem cells for 14d,and after that,immunofluorescence label and immunofluocytochemistry were used to observe the differentiation of neural stein cells. Result :MAP2-positive cells of LN group were more than others when LN was used as co-culture. GFAP-positive cells of poly-1-lysine group and matrigel group were more than that of LN group. Conelusion:LN promotes differentiation of neural stem ceils to neurons. LN is the best extracellular matrix for construction of the co-transplanted implant of neural stem ceils.
