摘要
目的探讨附子总生物碱对缺血心肌蛋白质组的影响。方法利用二维凝胶电泳分离左心室肌总蛋白,通过PDQuest 7.1.1软件分析2-DE图谱,比较和确定各组间差异蛋白的表达量,然后利用MALDI-TOF-MS制作差异蛋白点的肽谱确定各差异蛋白的种类,通过生物信息学分析所得蛋白的功能。结果附子总生物碱可调节缺血心肌的能量代谢、信号传导、机能、细胞修复和抗氧自由基损伤等多组相关蛋白的表达,对缺血心肌产生保护作用。结论附子总生物碱可调节缺血心肌多组蛋白表达,具有良好的保护缺血心肌作用。
Objective To reveal the mechanism of the anti-myocardial ischemia effect of total alkaloids from Radix Aconitine at the level of proteome. Methods The total proteins of left ventricular muscle were separated with 2-dimensional electrophoresis. The 2-DE images were analyzed with the PDQuest 7. 1.1 software package. The peptide mass fingerprinting was made with MALDI-TOF-MS, and then the identification and function of the protein were done via bioinformatics. Results Total alkaloids from Radix Aconitine could regulate the expression of 6 proteins in the ischemic myocardiurn. The proteins could be allocated into 5 groups: energy metablism-associated proteins, signal-associated proteins, self-repair-associated proteins, anti-free radical injury associated proteins, and myocardial contractile-associated proteins. Conclusion The total alkaloids from Radix Aconitine could prevent the myocardium from ischemia.
