摘要 目的观察5-羟色胺(5-HT)对丘脑底核(STN)神经元放电的影响,探索中缝背核(DRN)与STN的联系。方法应用细胞外记录的方法观察微电泳5-HT、赛庚定及氟西汀对STN神经元放电的影响。结果微电泳5- HT可使60.34%(35/58)STN神经元的放电频率降低(P<0.01);微电泳赛庚定使35.71%(10/28)神经元自发放电频率增加,多数神经元不受影响;但在微电泳5-HT过程中,给予赛庚定可使大多数(58.06%)神经元放电频率增加;氟西汀使56.25%(9/16)STN神经元放电频率降低。结论5-HT对STN神经元主要为抑制作用,赛庚定可拮抗其作用,氟西汀与5-HT作用效果一致。 Objective To research the effects of 5 - hydroxytrptamine (5 - HT) on the neuronal firing activities of the subthalamic nucleus (STN) in rats and to explore the relationship between the raphe dorsal nucleus (DRN) and STN. Methods Using extracellular recording to investigate the direct effects of iontophoretic applications of 5 - HT, cyproheptadine and fluoxetine on the spontanenous firing activities in STN. Results The averaged firing rate of 60.34% (35/58) STN neurons was decreased by 5 - HT (P 〈 0.01 ) , which was intensity - dependent. Majority of STN neurons were not responded to cyproheptadine, and cyproheptadine, while the spontaneous firing rate in 35.71% (10/28)of neurons was increased. During 5 - HT ejection, cyproheptadlne increased the spontaneous firing rate in majority(58.06% )of STN neurons. In addition, The averaged firing rate of 56.25% (9/16)STN neurons was decreased by fluoxetlne ( P 〈0.01 ). Conclusions STN neurons were inhibited by 5 - HT, cyproheptadine applied iontophoretically suppressed the inhibitory effects of 5 -HT, STN neurons were inhibited by fluoxetine as well as the effect of 5 -HT.
