摘要
目的 探讨电刺激丘脑底核对大鼠纹状体多巴胺能神经元递质释放的影响。方法 以深部电刺激术 (Deepbrainstimulation ,DBS)刺激 6 -OHDA损伤大鼠后 ,应用高效液相色谱 (highperformanceliquidchromatography ,HPLC)测定大鼠脑组织中的DA、Glu、GABA。结果 15 0Hz电刺激时 ,纹状体DA和GABA含量几乎达到正常对照组水平 ,与正常对照组差异无显著性 (10 .0 9± 1.4 7ng/mgvs 10 .6 5± 1.5 2ng/mg,P>0 .0 5 )。Glu的含量比较 ,在各种频率电刺激时 ,均显著低于正常组 ,但 15 0Hz电刺激组Glu含量较 5 0Hz、10 0Hz电刺激组有显著性升高 (2 .87± 0 .2 9μg/mgvs 2 .0 8±0 .2 2 μg/mg,2 .4 9± 0 .17μg /mg ,P <0 .0 1)。结论 DBS使纹状体多巴胺能神经元递质合成与分泌发生了改变。
Objective To explore the change of neurotransmitter of dopaminergic neuron in brain striatum during DBS-STN.Methods After 6-OHDA rats were stimulated by DBS, the concentration of DA, Glu and GABA was detected by HPLC.Results During 150Hz DBS, the concentration of DA and GABA had almost restored the level of the normal control group in brain striatum,and it had no significant difference compared with the normal control group (10.09±1.47ng/mg vs 10.65±1.52ng/mg, P>0.05). However, the concentration of Glu decreased compared with the normal control group in brain striatum during 50Hz, 100Hz and 150Hz DBS-STN, despite during 150Hz DBS-STN, the concentration of Glu had a significant increase compared with the 50Hz and 100Hz DBS-STN group (2.87±0.29μg/mg vs 2.08±0.22μg/mg,2.49±0.17μg /mg,P<0.01). Conclusions The synthesis and excretion of neurotransmitter of dopaminergic neuron in brain striatum had a change during DBS-STN.
