摘要
目的探讨orexin-B对正常大鼠丘脑底核神经元自发放电及运动行为的影响。方法应用在体电生理细胞外记录的方法观察orexin-B对大鼠丘脑底核神经元自发放电频率的影响,并采用提升躯体摇摆实验(EBST)观察丘脑底核微量注射orexin-B后大鼠运动行为随时间变化。结果大鼠丘脑底核微压力注射orexin-B,神经元放电频率平均升高(96.85±17.42)%(t=10.242,P<0.01),与生理盐水对照组的细胞反应百分数比较差异有极显著性(Z=-4.157,P<0.01)。EBST结果显示,单侧丘脑底核微量注射orexin-B引起大鼠向对侧摇摆,两组在观察的30、45s向对侧偏转的频率比较,差异有显著性(F=80.35,t=13.42、21.54,P<0.01)。结论 Orexin-B可提高正常大鼠丘脑底核神经元兴奋性,提示orexin-B可能通过基底神经节间接通路参与大鼠运动行为调控。
Objective To investigate the effects of orexin-B on spontaneous discharge and locomotive behavior in the subthalamic nucleus of normal rats. Methods Applying in vivo electrophysiological extracellular recording method,the effect of orexin-B on the spontaneous discharge frequency of rat subthalamic nucleus neurons was observed,and adopting elevated body swing tests(EBST),the changes of motor behavior in rats with time after microinjection of orexin-B into the subthalamic nucleus were observed. Results After micro-pressure injection of orexin-B into rat subthalamic nucleus,neuron discharge average increase of(96.85±17.42)%(t=10.242,P〈0.01),compared with normal saline group,the difference in percentage of cell response was significant(Z=-4.157,P〈0.01).EBST results showed that unilateral subthalamic nucleus microinjection of orexin-B caused the rats to swing to the opposite site.The difference in frequency of deflection to the opposite site-in 30 and 45s of observation-between the two groups was significant(F=80.35;t=13.42,21.54;P〈0.01). Conclusion Orexin-B can increase the excitability of subthalamic nucleus neurons in normal rats,which suggests that orexin-B involves in regulation of motor behavior in rats through indirect pathway of basal ganglia.
