乙肝疫苗接种后无弱应答者发生的分子生物学机制(英文) 被引量:7

摘要 目的研究乙肝疫苗接种后无弱应答的发生与IL-2或IL-6水平之间的关系,从分子生物学角度探讨无弱应答者发生的分子生物学机制。方法从健康人群中随机选取30个乙肝疫苗接种后无弱应答者和正常应答者作为本次研究的对象。采用CTLL-2和7TD1方法监测IL-2和IL-6活性水平。比较两组IL-2和IL-6活性水平,并建立起IL-2和IL-6活性水平与抗-HBs滴度之间的回归方程,以通过IL-2和IL-6活性水平来预测乙肝疫苗接种后无弱应答的发生。结果乙肝疫苗接种后无弱应答的IL-2活性水平的平均值明显低于正常应答者(P<0.01),无弱应答的IL-2活性水平和正常应答者的平均值分别为74.5±64.1u/ml和298.0±101.1u/ml。并且相关回归分析发现,在乙肝疫苗接种后正常应答者组中抗-HBs滴度与IL-2活性水平之间呈明显正相关,相关系数为r=0.68。正常应答者组的IL-6活性水平(58.98±16.4)明显高于无弱应答者组(30.10±12.4)(P<0.05),而且在乙肝疫苗接种后正常应答者组中抗-HBs滴度也与IL-6活性水平之间呈明显正相关,相关系数为0.79(P<0.05)。结论乙肝疫苗接种后无弱应答者的发生可能与被接种者体内IL-2、IL-6活性水平有关。提示我们,为了降低乙肝疫苗接种后无弱应答者的发生率,可以在注射乙肝疫苗的同时注射少量IL-2或IL-6。 Objective AIM:To explore the relationship between non- and- hyporesponders to hepatitis B vaccine and their serum interleukine - 2 ( IL - 2) or interleukine - 6 (IL - 6) levels , Methods Random selected 30 non- and - hyporesponders and 30 normal responders to HB vaccine from healthy people, these were regard as researched subjects. Used CTLL - 2 and 7TD1 method to determination IL - 2 and IL - 6 activity leveIs, Compared to IL- 2 and IL - 6 activity leveIs between two groups , and building regression equation between IL - 2 or IL- 6 activity leveIs an d anti- HBs titres. Results IL- 2 meanl activity levels of the non- and- hyporesponders to hepatitis B vaccine is much less than IL - 2 mean activity of the normal responder to hepatitis B vaccine(P〈0.01) ,the IL- 2 mean activity levels of the non- and- hyporesponder groups and the normal responder groups were 74.5 ± 64, 1 u/ml and 298.0 ± 101. 1u/ml respectively. The relationship between the anti - HBs levels of normal response to hepatitis B vaccine and IL - 2 activity levels was 0.68 (r = 0.68 P 〈 0.05) . Interleukin- 6 mean activity levels of the responder groups(58.98± 16, 4)had a significantly higher than that the non - and - hyporesponder groups (30.1 0 ± 1 2.4) (P 〈 0.05), there was significant positive correlation between the IL- 6 activity levels and levels of titers of serum anti - HBs in the responder groups, the correlation coefficents was 0. 79(P 〈 0. 05). Conclusion There was significant positive correlation between the IL - 2 or IL- 6 activity levels and levels of titers of serum ami - HBs. For enhance non - and - hyporesponders rate to hepatitis B vaccine , we can injected some cytokines as IL - 2 or IL - 6 when injected HB vaccine.
