当归注射液影响辐射损伤小鼠骨髓基质细胞血管内皮生长因子及其受体flt-1的表达 被引量:7

Influences of angelica sinensis injection on the expressions of vascular endothelial growth factor and Flt-1 in murine bone marrow stromal cells following radiation injury

摘要 目的:观察当归注射液对辐射损伤小鼠骨髓基质细胞血管内皮生长因子及其受体Flt-1表达的影响。方法:实验于2005-08/2005-11在武汉大学人民医院儿科研究所进行。将56只健康BALB/c小鼠随机分为3组:正常组:8只小鼠不做处理;照射组:24只小鼠经60Coγ射线6.0Gy全身照射;当归注射液组:24只小鼠经60Coγ射线照射后给予腹腔注射当归注射液2.5g/(kg·d)。在照射后第3,7,14天采用免疫组化法检测小鼠骨髓基质细胞血管内皮生长因子及其受体Flt-1的表达,并用流式细胞术检测相应时间的骨髓基质细胞凋亡率。结果:56只小鼠均进入结果分析。①当归注射液组及照射组骨髓有核细胞计数均低于正常组,当归注射液组照射后7,14d骨髓有核细胞计数高于照射组[正常组:(7.9±0.9)×106/根股骨;照射组:(1.3±1.0)×106/根股骨,(1.8±1.1)×106/根股骨,(2.1±0.7)×106/根股骨;当归注射液组:(1.6±0.9)×106/根股骨,(4.6±1.2)×106/根股骨,(5.6±0.9)×106/根股骨,P<0.05或P<0.01];当归注射液组照射后7,14d血管内皮生长因子及其Flt-1受体表达明显高于照射组[当归注射液组:(122.5±1.0),(128.0±0.9),(139.2±0.8),(146.8±0.8)μm3;照射组:(112.6±1.0),(108.4±0.9),(124.4±1.3),(132.5±1.1)μm3,P<0.05或P<0.01]。②照射组和当归注射液组骨髓基质细胞凋亡率明显高于正常组,当归注射液组照射后3,7,14d骨髓基质细胞凋亡率均明显低于照射组[正常组:(2.2±1.0)%;照射组:(18.6±3.4)%,(17.6±1.5)%,(11.4±3.7)%;当归注射液组:(13.8±1.3)%,(11.6±4.7)%,(6.3±2.7)%,P<0.05或P<0.01]。结论:当归注射液可能通过促进骨髓基质细胞血管内皮生长因子及其受体表达,修复骨髓微环境。 AIM: To investigate the influences of angelica sinensis injection on the expression of vascular endothelial growth factor (VEGF) and its receptor Fit-1 in murine bone marrow stromal cells with radiation injury. METHODS: The experiment was carried out in the Institute of Pediatrics, Renmin Hospital, Wuhan University from August to November 2005, The 56 BALB/c mice were randomly divided into 3 groups: Group A was normal control group with 8 mice (did not received any dispose), Group B as radiation group with 24 mice had been generally irradiated by ^60Co 6.0 Gy γ-ray, Group C as angelica sinensis injection group with 24 mice was intraperitoneally injected with angelica sinensis injection 2.5 g/kg per day after irradiated. On days 3, 7 and 14 after irradiation, the expressions of VEGF and Fit-1 in murine bone marrow stromal cells were assayed by immunohistochemical method. The apoptosis rate of bone marrow stromal cells was tested by flow cytometry. RESULTS: Totally 56 mice were involved in the result analysis. ①Bone marrow nucleated cells (BMNCs) were less in the Group C and Group B than Group A. On days 7 and 14 after radiation, the BMNCs were more in the Group C than Group B [Group A: (7.9±0,9)×10^6 femur;Group B: (1,3±1.0)×10^6/femur, (1,8±1.1)×10^6/femur, (2,1±0.7)×10^6 femur; Group C: (1.6±0.9)×10^6/femur, (4,6±1.2)×10^6 femur, (5,6±0,9)×10^6/femur,P 〈 0,05 or P 〈 0.01], On the days 7 and 14 after radiation, expression of VEGF and Fit-1 in Group C were significantly higher than that in the Group B [Group C: (122.5±1.0), (128.0±0.9), (139,2±00,8), (146,8±0.8)μm^3;Group B: (112,6±1,0), (108,4±0.9), (124,4±1.3), (132.5±1.1)μm^3, P 〈 0.05 or P 〈 0,01]. ②The apoptotic rate of bone marrow stromal cells were obviously higher in the Group B and Group C than the Group A, while after 3, 7 and 14 days radiation the apoptotic rate was markedly lower in the Group C than the Group B [Group A: (2
