摘要
目的观察血管性痴呆中Tau蛋白表达和磷酸化程度,研究其在发病机制中的可能作用。采用持久性双侧颈总动脉结扎方法制备血管性痴呆模型,Morris水迷宫检测大鼠空间学习记忆能力的差异,HE染色和免疫组化染色检测海马CA1区Tau蛋白、蛋白磷酸酯酶2A的表达情况。结果发现手术组与对照组大鼠相比,Morris水迷宫隐藏平台逃避潜伏期延长,空间摸索试验时间减少;海马CA1区细胞排列疏松,数目减少,细胞形态异常;手术组大鼠神经元中Tau蛋白含量增多,蛋白磷酸酯酶2A减少,以一个月时最明显。以上结果表明Tau蛋白表达增多及过度磷酸化在血管性痴呆发病机制中可能起到作用。
Objectlve The present study was aimed to investigate the role of Tau protein on the mechanism of vescuhr dementia. Fifty Wistar rats weighted from 300 g to 400 g were randondy divided into the control group( n = 10)and the operation group( n = 40), and the latter gsoup were equally divided into four groups according to the operation time: 1, 2,3 and 4 month group. Tje bilsteral common carotid arteries of the experimental mrs were ligated so as to construct a model of vascular deanentia rat. The abilities of learning and memorizing of mrs were tested by using the Morris water maze. The rats were trained in Morris water maze program for five days to find the escape latencies and the times of spatial probe test. HE stain was used to obverse the morphology of neurofibrils in CA1 subfield of hippocampus.lmmunohistechemistry staining was used to obverse Tau protein and protein phosphatase 2A in CA1 subfield of hippocampus. Compared with the control group, The escape latencies were significant longer and the times of spatial probe test was decreased in operation group.The HE stain of operation group showed the pyramidal cells were lost,degenerative and necrotic in quantity,some nuclei were stained deeply and pynotic, and the dendrite of the top was prolonged.The immunohistochemistry results showed a significant increase in phosphorylation of Tau protein and decrease of protein phosphatase 2A in the operation group relation uo the control group,especially in 1 month and 2month group.These results indicated the phosphorylation of Tau protein play an important role on the mechanim of vascular dementia. These data also implicate that the role of Tau protein maybo have some relationship with apoptosis.
