电针肾俞穴对吗啡慢性注射大鼠学习记忆的影响 被引量:5

摘要 目的建立大鼠吗啡慢性注射学习记忆损害模型,观察电针肾俞穴对模型大鼠学习记忆的改善作用,探讨电针对吗啡慢性注射大鼠学习记忆损害的防治作用。方法通过慢性皮下注射吗啡(10mg/kg,30mg/kg)21d,建立SD大鼠吗啡慢性注射学习记忆损害模型。采用Morris水迷宫装置,记录大鼠上平台时间(潜伏期)及寻找平台路线(游泳总距离),观察电针对吗啡慢性注射大鼠学习记忆的恢复作用。结果第12天吗啡中剂量组Morris水迷宫测试上平台时间[(74.00±19.26)s]明显延长,与生理盐水组[(32.50±18.86)s]、东莨菪碱组[(24.66±8.73)s]相比,差异有显著性(P<0.05)。在第18天,第20天,电针治疗组大鼠上平台时间[(13.75±3.63)s],游泳总距离[(168.47±119.43)cm],与吗啡组[(81.00±17.58)s和(469.71±268.72)cm]相比,差异具有显著性(P<0.05)。结论吗啡慢性注射可以导致大鼠学习记忆损害,而电针对大鼠吗啡慢性注射所造成学习记忆损害有一定的改善作用。 Objective To build up the model of morphine chronic injection with learning and memory lesiorl model in rats,and to observe the effect of electro-acupuncture ( EA ) at the acupuncture point Shen-Shu on morphine-induced rats model, and to analyze the preventive action of EA on chronic injection morphine rats. Methods Chronic injection morphine model in rats was established by chronic subcutaneously (so.) injection morphine (10,30mg/kg) 21days. The time of reaching platform and its lime of reaching platform and the improve action of EA on morphine-induced lesion of learning-memory in rats were recorded by Morris water maze test. Resuits During Morris water maze test at 12th day, the time of reaching platform was longer in middle-dose morphine(10mg/kg) group(74.00 ± 19.26)s than that of saline group (32.50 ± 18.86)s and that of scopolamine gronp( 24.66 ± 8.73 ) s. Meantime, its time of reaching platform was ( 81.00 ± 43.06 ) s and that of saline group was ( 13.75 ± 7.27 ) s ( P 〈 0.05 ). The swimming distance was ( 168.47 ± 119.43 ) cm and (469.71 ± 268.72) cm, respeetively, and it appeared significant difference, compared with that in middle-dose morphine group. Conclusion Chronic injection morphine could induee defects of the learning and memory in the rat. Meantime, EA could improve morphine-induced lesion of learning-memory.
