摘要
目的:建立眼眶腺样囊性癌裸鼠模型,通过瘤内注射化疗药物使肿瘤内药物达到高浓度,确定最佳药物浓度,从而减少大剂量化疗药物对全身损害,为局部注射化疗药物治疗眼眶恶性肿瘤提供动物实验基础和临床参考。方法:通过瘤细胞悬液接种法建立裸鼠眼眶原位移植腺样囊性癌模型;比较全身化疗和局部化疗对肿瘤增殖的抑制作用,得出局部化疗最佳药物浓度;电镜和光镜观察,初步研究局部化疗的副作用。结果:接种瘤细胞7d后,37只裸鼠成瘤,成瘤率100%;局部注射VCR2μg时,与全身用药的抑瘤率和治疗效果无显著性差异(77.82%,66.65%;P>0.05),当注射VCR3μg时,其抑瘤率与全身用药组相比有显著性差异(93.09%,66.65%;P<0.05),当注射VCR4μg时,其抑瘤率与全身用药组相比,结果无显著性差异(82.82%,66.65%;P>0.05);局部注射区皮肤及眶内软组织病理组织学检查,均未见局灶性坏死等副作用。结论:采用瘤细胞悬液接种法成功建立裸鼠眼眶腺样囊性癌动物模型;局部注射VCR的最佳治疗浓度为0.03g/L;注射局部未见毒副反应,瘤内注射是一种安全有效的化疗方法。
AIM: To reduce the systemic side effects and provide experimental basis and clinical references for local chemotherapy on orbital malignant tumors by building nude mouse model of orbital adenoid cystic carcinoma (ACC) and determining the effective drug concentration.
METHODS: Orthotopic nude mouse model of orbital ACC was built by injecting ACC cell suspension. The effective drug concentration was determined by comparing systemic chemotherapy and local chemotherapy. The side effects of loCel chemotherapy were studied by observation of electric microscope and light microscope. RESULTS: The ACC model was succeeded in all nude mouse in 7 days. The tumor inhibition rate of VCR 2μg, VCR 3μg, VCR 4μg and systemic chemotherapy was 77.82%, 93.09%, 82.82% and 66.65%, respectively. The difference between VCR 3μg and systemic chemotherapy exhibited statistical significant difference (P〈0.05). The histological examination of skin and parenchyma in the mice orbit of local chemotherapy showed no pathologic changes. CONCLUSION: Orthotopic nude mouse model of orbital ACC was built by injecting ACC Cell suspension. The best VCR concentration for intralesional injection was 0.03g/L. No side effect was observed and it would be one of the effective methods of chemotherapy for orbital ACC.
