摘要
目的探讨高频超声在裸鼠前列腺原位种植肿瘤模型中的应用价值。方法建立裸鼠前列腺原位种植肿瘤模型42只。肿瘤接种后分别在第2~8周,随机抽取6只裸鼠行高频超声检查,观察肿瘤的形态、回声、边界,测量肿瘤大小,彩色多普勒超声观察肿瘤的血流分布情况。每次超声检查后,立即麻醉、解剖裸鼠。分离出前列腺肿瘤组织.卡尺测量肿瘤的大小,而后行HE染色。分析超声测量结果与卡尺测量结果的相关性。结果高频超声与游标卡尺测量结果呈高度正相关(r=0.996,P<0.001)。超声观察前列腺种植肿瘤呈椭圆形,边缘规则,中低回声,2~3周肿瘤体积变化很小,4~6周肿瘤生长最快。彩色血流信号最丰富;7~8周生长减慢,内部出现无回声的液化坏死区,彩色血流信号减少,与周围边界不清。结论高频超声能准确监测裸鼠前列腺原位种植肿瘤模型的生长,有较强的实用价值。
Objective To assess the value of high frequency ultrasound in monitoring the growth of orthotopic prostate tumor in nude mice model.Methods Forty-two nude mice were implanted with prostate tumor cell suspensions.Six mice were chosen randomly and the size,internal echo and blood supply of tumor were observed by high frequency ultrasound at 2-8 weeks after the first injection.After the examination,the nude mice were dissected under narcotics and the tumors were dissociated immediately.The transversal and sagittal diameter were measured by vernier caliper.Tumors were fixed for HE staining.Results The volume of tumor measured by high frequency ultrasound and vernier caliper had strong correla- tion(r=0.996,P〈0.001).High frequency ultrasound could also observe resonance,blood supply and necrosis of the tumors.The fast period of tumor growth with the richest blood supplying appeared 4-6 weeks after injection.Conclusion High frequency ultrasound provides an accurate,practical method to monitor the growth of orthotopic prostate tumor in nude mice model.
