摘要
本实验对切除颌下腺35天、52天和70天的小鼠做了睾丸生精细胞DNA含量的流式细胞分析.结果表明:①各实验组的1C细胞群(精子细胞和精子)和4C细胞群(初级精母细胞)比例明显低于相应的对照组(P<0.01),2C细胞群(精原细胞和次级精母细胞等)比例明显高于对照组(P<0.01).②实验组各峰的均道值低于对照组(P<0.01),即DNA含量低于对照组.③细胞周期分析显示实验组的G0/G1期和S期细胞比例增加(P<0.01),G2/M期细胞比例明显减少(P<0.01),同时PI值明显降低(P<0.01)。上述结果提示体内表皮生长因子(Epidermelgrowthfactor,EGF)缺乏可导致小鼠睾丸初级精母细胞和精子细胞减少,并可影响细胞的DNA合成和延缓生精细胞的增殖.
Abstract The DNA content of testiular spermagonic cells of male mice with epidermal growth factor (EGF) deficiency caused by sialoadenectomy for 35, 52 and 70 days was examined by flow cytometry. The rsults showed: ①proportion of 1C cells (spermid and sperm) and 4C cells (primary spermatocyte) in experimental groups were significantly lower than those in corresponding control groups (P < 0. 01 ), and proportions of 2C cells (spermatogonia and secondary spermtocyte) in experimental groups were markedly higher than those in the control groups (P<0. 01) ;②average channel values of all peaks in experimental groups were lower than those in the control groups (P<0. 01 ),meaning the DNA contents were lower than those in the control groups;③ percentage of cells G0/G1 phase and S phase in experimental groups were increased, G2/M phase were reduced, as well as PI (P<0. 01). These findings suggest that EGF deficiency in mice may result in a reduction of primary spermatocyte and spermatid, influence the synthesis of DNA in the cells, and produce a delay in proliferation of spermatogenic cell.
