摘要
目的优化基因工程菌Escherichia coli BL 21生产人源型胶原蛋白的高密度发酵工艺。方法建立一套以溶解氧浓度和比生长速率为中心的控制策略,采用溶氧反馈或pH反馈调节补料速度。结果 OD600可达150,胶原蛋白的表达量为9g/L。结论 20%-30%饱和度的溶解氧和0.1- 0.2 h-1的比生长速率有利于细胞生长和产物表达,其与降低乙酸等有害代谢副产物的积累密切相关。
Aim To study and optimize the process of the high cell-density fermentation of recombinant E. coli BL 21 and the expression of the human-like collagen. Methods A strategy in which the dissolved oxygen (DO) and specific growth rate (μ) were as the key parameters was applied. The glucose feeding control was DO and pH- stats, which is aimed to suppress the metabolic activity of acetic acid synthesis. Results Finally, the A600 ( OD600 ) value reached 150 and the collagen with a yield of 9 g/L was obtained. Conclusion It was beneficial to the cell growth and aim protein expression when the dissolved oxygen was controlled at 20% -30% and the specific growth rate at 0.1 -0.2 h^-1. And it could reduce the accumulation of acetic acid.
