摘要
目的:观察使用苦参碱预防兔耳创面增生性瘢痕形成的作用。方法:实验于2004-04/11在南方医科大学附属珠江医院心内科实验室及病理科完成。在10只兔耳作80个创面建立增生性瘢痕的动物模型。随机分为苦参碱组和生理盐水组,每组5只兔,40个创面。在做兔耳创面后的第2天起,开始使用药物,每次取0.5m L液体点至创面,3次/d。苦参碱组使用苦参碱贮液,生理盐水组使用生理盐水。在第3周切除兔两耳增生性瘢痕,做苏木精-伊红染色、M asson染色,苦味酸天狼猩红染色。观察两组兔耳创面中不形成增生性瘢痕的创面愈合时间及形成增生性瘢痕的数量;胶原纤维的面密度比、Ⅰ型和Ⅲ型胶原的面密度比值。结果:10只大白兔、80个创面均进入结果分析。①大体观察:苦参碱组未形成增生性瘢痕有12个创面(30%),平均愈合时间为(10.6±1.7)d,形成增生性瘢痕块有28个创面(70%)。生理盐水组未形成增生性瘢痕有9个创面(22.%),平均愈合时间为(10.1±1.1)d,形成的增生性瘢痕块有31个创面(77.5%),两组间无统计学差异(P>0.05)。②光镜观察:苦参碱组平均胶原纤维面密度比明显较生理盐水组低(0.481±0.048,0.604±0.040,P<0.001);苦参碱组平均Ⅰ/Ⅲ型胶原的面密度比值显著高于生理盐水组(2.574±1.006,2.286±0.416,P<0.001)。结论:苦参碱不影响兔耳创面的正常愈合,也不能预防兔耳创面增生性瘢痕的形成,但可以减轻所形成的增生性瘢痕的增生程度。
AIM: To investigate the effect of matrine on preventing the formation of hyperplastic scar of wound of rabbit ears. METHODS: The experiment was conducted at the laboratory of Department of Cardiology, Department of Pathology, Affiliated Zhujiang Hospital, Southern Medical University from April to November 2004. Animal models with hyperplastic scar in 10 rabbit ears having 80 wounds were established. The rabbits were divided into matrine group and saline group randomly, each group having 5 rabbits with 40 wounds totally. From the seeond day of operation on the rabbit ears, drugs were began to adopt, 0.5 mL liquid was dropped on raw surfaces every time, 3 times per day. Those in the matrine group were treated with matrine, and those in the saline group were given with saline. The hyperplastic scars were cut out after three weeks, and made in HE stain, Masson stain, and pictric acid-sims red stain respectively. The heal time of normal heal scars, the numbers of hyperplastic scar, the collagenous density and Ⅰ/Ⅲ type collagenous density on area were observed, RESULTS: Ten rabbits and 80 wounds were all involved in the result analysis. ① General observation: Normal healed scars in the marime group were 12 (30%), and average healed time were (10,6+l,7) days. There were 28 wounds with hyperplastic scar (70%), 9 scars were normal heal in the saline group (22.%), and the average heal time were (10.l+l.l) days. There were 31 wounds with hyperplastic scar (77.5%).There was no statistical significance between the two groups (P〉0.05). ②Observation with electron microscope: The average collagenous density on area of matrine group was significantly lower than that in the saline group (0.481±0.048, 0.604±0.040 ,P〈0.001 ). The average Ⅰ/Ⅲ type collagenous density on area of matrine group was higher obviously than that in the Saline group ( 2.574±1.006,2.286±0.416, P〈0.001 ). CONCLUSION: The applied matrine cannot either affect the normal healing of wound of
