昆虫病原线由固体培养系统干粉培养基的优化 被引量:2

OPTIMIZATION OF THE MEDIUM COMPONENTS FOR THE SOLID CULTURE OF ENTOMOPATHOGENIC STEINERNEMA AND HET ERORHABDITIS NEMATODES

摘要 本文根据系统分析方法,利用二次正交旋转组合设计于固体培养系统中建立培养基组份对S.carpocapsae的A24品系和H.bacteriophora的H06品系的线虫产量影响的数学模型,然后对模型进行多项分析和优化。 根据营养配比适合、来源便利、质量稳定以及成本适宜的原则,选择黄豆粉、面粉、鸡蛋黄粉、猪油以及酵母膏作为线虫固体培养基组份。水份、酵母膏和鸡蛋黄粉对A24线虫产量产生显著的影响;水份、猪油对H06线虫产量产生显著影响。对A24和H06线虫来说,不同组份的重要性程度不同,表明根据不同的线虫种来筛选合适的培养基组份及其配比是极为必要的。在36个试验组合中,A24和H06线虫最高产量的组合分别获得每克培养基58×10~4和48×10~4条的感染期线虫。通过对两种线虫的培养基配比模型进行优化,得到了优化的培养基配方。为使优化配方更有广泛性,以频数分析方法进一步求得一定置信区间(95%)内各培养基组份的最佳配比范围。 The rotatable composite design was introduced to determine the effects or interac-tions of medium compositions (soy flour, wheat flour, yeast extract, egg yolk, lard and water added) on the growth and yield of Steinernema carpocapsae strain A24 and Het-erorhabditis bacteriophora strain H06 in Bedding's solid culture system.Nematode yields were significantly affected by water added, yeast extract and egg yolk for A24, and by water added and lard for H06, which indicated the importance of selecting suitable medium components and combinations for a said nematode strain. The highest yield of 58 ×104 per g medium from A24 and 48×104 per g medium from H06 were obtained. Optimum combinations of medium components (with 95% confidence limits) for both strains were computed and confirmed by the replicated tests.
