摘要
[目的]研究苯并(a)芘(BaP)、铅单独及其联合作用对小鼠大脑神经元DNA的损伤。[方法]将80只昆明种小鼠随机分为10组(每组8只) ,即空白对照组、溶剂对照组、低浓度铅染毒组、高浓度铅染毒组、低剂量BaP染毒组、高剂量BaP染毒组、低浓度铅+低剂量BaP联合染毒组、低浓度铅+高剂量BaP联合染毒组、高浓度铅+低剂量BaP联合染毒组及高浓度铅+高剂量BaP联合染毒组。空白对照组不作处理,溶剂对照组用等容量植物油平行处理;低、高浓度的铅染毒分别为5 .4、5 4mg/L的醋酸铅饮水染毒;低、高剂量BaP染毒分别为0 .5、5mg/kg的BaP植物油溶液,每周4次腹腔注射,联合染毒组接受两种处理。染毒8周后取各组小鼠脑组织制作细胞悬液,单细胞凝胶电泳法检测小鼠大脑神经元DNA的损伤。[结果]①单独染毒:BaP 5mg/kg染毒组小鼠大脑神经元DNA的损伤程度高于对照组(P <0 .0 5 )。②联合染毒:5 .4mg/L的醋酸铅+ 0 .5mg/kg的BaP染毒组(P <0 .0 5 ) ;5 .4mg/L的醋酸铅+ 5mg/kg的BaP染毒组(P <0 0 1) ;5 4mg/L的醋酸铅+ 0 .5mg/kg的BaP染毒组(P <0 .0 1) ;5 4mg/L的醋酸铅+ 5mg/kg的BaP染毒组(P <0 .0 1)小鼠大脑神经元DNA的损伤程度显著高于对照组。[结论]高剂量BaP连续8周暴露可损伤小鼠大脑神经元;
Objective] To study on the neuron DNA damage induced by benzo[a]pyrene or lead alone and in combination in mice. [Methods] 80 male Kunming mice were randomly divided into 10 groups and 8 animals per group were used,they were ①control group;②vehicle group;③low dose lead group;④high dose lead group;⑤low dose BaP group;⑥high dose BaP group;⑦low dose BaP+low dose lead group;⑧low dose lead+high dose BaP group;⑨high dose lead + low dose BaP group;B10high dose lead+high dose BaP group.The control animals were without any treatment,and the vehicle group of mice received an equal volume of corn oil only.Animals in low and high dose lead groups were administrated PbAc through drinking water at concentrations of 5.4 or 54 mg/L while animals in low and high dose BaP groups were intraperitoneally administrated BaP dissolved in corn oil at the dose levels of 0.5 or 5.0 mg/kg,4 times per week.Therefore animals in combine-treated groups received two treatments simultaneously.On the end of 8-week administration,all mice were killed and the brains were removed.Then cracks on DNA straps in the mice brain neurons were detected by comet assay. [Results] ①Single administration:The DNA damages on the brain neurons in 5 mg/kg BaP mice were significantly serious than that of control mice(P<0.05).②Combine administration:The DNA damages on the brain neurons in 5.4 mg/L PbAc+0.5 mg/kg BaP mice(P<0.05);5.4 mg/L PbAc+5 mg/kg BaP mice(P<0.01);54 mg/L PbAc+0.5 mg/kg BaP mice(P<0.01);54 mg/L PbAc+5 mg/kg BaP mice(P<0.01) were significantly serious than that of control mice. [Conclusion] Exposure with high dose BaP can damage CNS neurons on mammals;damages of BaP on mice brain neurons can be enhanced by lead.
