干扰素-γ对单核—巨噬细胞源性泡沫细胞ACAT-1表达的影响 被引量:8

摘要 目的观察干扰素-γ对THP-1、THP-1源性巨噬细胞泡沫细胞酰基辅酶A胆固醇酰基转移酶-1(ACAT-1)表达的影响。方法将THP-1细胞与PMA孵育48小时使之分化为巨噬细胞,继以100mg/mlOx-LDL处理24小时使之形成泡沫细胞。将THP-1细胞、巨噬细胞、泡沫细胞分别用300U/mlIFN-γ干预24小时。RT-PCR检测ACAT-1mRNA水平,WesternBlot检测其蛋白表达。结果THP-1细胞分化成巨噬细胞以及形成泡沫细胞时ACAT-1mRNA及蛋白含量增加(P<0.05),而后两种细胞之间无显著性差异。与各自的对照组相比,经IFN-γ作用后三种细胞ACAT-1mRNA及蛋白表达均增强(P<0.05)。结论IFN-γ诱导单核、巨噬细胞、泡沫细胞ACAT-1的mRNA及蛋白表达,这可能是其促进动脉粥样硬化的机制之一。 Objective To investigate the role of interferon-γ (IFN-γ ) in acyl-coenzyme A cholesterol acyltransferase-1 (ACAT-1) expression in THP-1 and THP-1-derived macrophages and foam cells.Methods THP-1 cells were incubated with PMA for 48 hours then exposed to 100 mg/ml Ox-LDL for 24 hours,in the presence or absence of IFN-γ.The ACAT-1 mRNA and protein levels were detected by RT-PCR and Western Blot respectively.Results IFN-γ enhanced significantly ACAT-1 mRNA and protein levels in THP-1 and THP-1-derived macrophages and foam cells (P<0.05).Conclusion IFN-γ up-regulates ACAT-1 expression in monocyte-macrophage and foam cells,which may be involved in the proatherogenic effects of IFN-γ.
